Once the Nazis assumed control in 1933, Hess was named Deputy Fuhrer, a position that placed him second only to Hitler. While he remained loyal and dedicated, his role was largely supportive rather than authoritative, particularly when compared to figures like Heinrich Himmler, who built a substantial power base within the regime. Nevertheless, Hess was deeply involved in crafting the regime's policies, including the Nuremberg Laws, which stripped Jews of their rights.

As World War II commenced, Hess’s focus began to shift. He took a position on the war cabinet but found himself increasingly sidelined by Martin Bormann, who cultivated a deep power base at Hess's expense.
